You can look at the donor bike how the engine is mounted - in rubbers or not.
RD 400 has rubber mountings and a cushion drive.
It's a question of torque peaks. The fewer cylinders, the larger the displacement, the less distributed the ignition points are and the less crankshaft mass, the greater the torque peaks. It's about the gear wheels, chain and sprockets and also the rear tires (but there only about wear). Although without the experience of no cushion drive I would use one, if there is the chance.
Noise could be another aspect.
